Title: Impact of rootstock on Vitis response to drought

Abstract: Drought stress affects yield and berry quality in grapevine. Different vine species and varieties are capable to cope with water stress to very different degrees, from extremely sensitive to more tolerant. The use of interspecific hybrids as rootstocks can determine the response to stress. However, the mechanisms involved in this process are not fully known. With the VitiRoots project, we aim to characterize the impact of rootstocks in grapevine at physiological and molecular level in response to water stress. Castelão (Vitis vinifera) was used as the scion for both grafts, and Paulsen 1103 (Vitis berlandieri x Vitis rupestris) and SO4 (Vitis berlandieri x Vitis riparia) as the rootstocks, that have been described as more tolerant and more susceptible to water stress, respectively. microRNAs are, including drought.
We used a physiological approach to assess and characterise plant responses to water stress. At the molecular level, we have given particular focus to the microRNAs–small non-coding RNAs that negatively regulate target genes at post-transcriptional level in response to abiotic stresses –differentially expressed in response to drought.
Here, we present preliminary data on the miRNAome in leaves and roots of the two grafted grapevines in response to water deficit conditions.
