Title: Large scale MS-based proteomics for global drug profiling and targeting proteostasis

Abstract:
Large scale MS-based proteomics have a broad range of applications in biology and health science. MS-based proteomics have over the last decades been optimized to allow profiling of a number of post translational modifications on large scale such as phosphorylation, acetylation and glycosylation. However, there are more than 200 known protein modifications leaving plenty of undiscovered territory to be explored. Furthermore, post translational modifications are often deregulated in disease conditions which have been explored for biomarker application but also in several therapeutic strategies.
The current state of art proteomics can provide a fairly complete quantitative overview of a cellular or tissue proteome at cost effectiveness similar to transcription analysis. This opens the possibility to use MS for pharmacodynamics in addition to the frequent use of MS for biomarker discovery. The main part of this presentation is devoted to discuss our experience and results obtained by using state of art MS for global drug profiling. However, examples of the use of large scale MS in proteostasis research from current project collaboration within the COST action PROTEOSTASIS network will be highlighted.
