Technical expertise

I use an integrated structural biology approach to improve our knowledge about DNA repair and molecular adaptation to extreme environment. This includes: 

- Cloning, small and large scale expression (fermentation) and purification of recombinant enzymes (includes bioinformatic analysis of protein targets prior to production).

- Biochemical and biophysical characterisation of recombinant enzymes (enzyme assays to determine activity, specificity and calculate kinetic constants, thermofluor, differential scanning calorimetry (DSC), isothermal titration calorimetry (ITC) and surface plasmon resonance (SPR).

- Electrochemical (cyclic voltammetry) and spectroscopic analysis (UV-visible, EPR and resonance raman) of recombinant enzymes holding metal centers.

- Protein crystallisation, optimisation and structure determination by X-ray crystallography.
